چکیده

background tyrosinase is a copper-containing enzyme, which is widely distributed in microorganisms, animals and plants. it is also a key enzyme in melanin biosynthesis, which plays a crucial role in determining the color of mammalian skin and hair. in addition, unfavorable enzymatic browning of plant-derived foods by tyrosinase causes a decrease in nutritional quality and economic loss of food products. objectives in the present study the activity of this enzyme was examined against quersetin and kaempferol as two potentially flavonoid inhibitors. materials and methods in this work, the effects of quercetin and kaempferol as propolis-derived compounds on activity of mushroom tyrosinase (mt) were studied. these flavonoids showed inhibitory activity on catecholase and cresolase reactions in presence of caffeic acid and p-comaric acid, respectively. the inhibition mode of quercetin and kaempferol were competitive towards both catecholase and cresolase activities of the enzyme. results the inhibition constants (ki) were determined as 0.072 and 0.112 mm for catecholase activity, and 0.016 and 0.06 mm for cresolase activity, respectively. conclusions in general, quercetin and kaempferol can be used as good candidates in melanogenesis inhibition. moreover they should be considered as good blockers of enzyme activity in hyper pigmentation and clinical application.

In vitro and in silico studies of the inhibitory effects of some novel kojic acid derivatives on tyrosinase enzyme

Objective(s): Tyrosinase is a key enzyme in pigment synthesis. Overproduction of melanin in parts of the skin results in hyperpigmentation diseases. This enzyme is also responsible for the enzymatic browning in fruits and vegetables. Thus, its inhibitors are of great importance in the medical, cosmetic and agricultural fields. Materials and Methods: A series of twelve kojic acid derivatives wer...

Tyrosinase inhibitory prenylated flavonoids from Sophora flavescens.

For the purpose of the development of a skin-whitening agent, Sophora flavescens was evaluated for tyrosinase inhibitory activity and its active principles were identified following activity-guided isolation. The ethanol extract and dichloromethane fraction from S. flavescens showed significant inhibition of mushroom tyrosinase. From the dichloromethane fraction, three known prenylated flavonoi...

Tyrosinase inhibitory properties of phenylpropanoid glycosides and flavonoids from Teucrium polium L. var. gnaphalodes

Objective(s): In food industry, the inhibition of tyrosinase is very important, because this enzyme catalyzes the oxidation of phenolic compounds found in fruits and vegetables into quinones, which contribute in undesirable color and taste of fruits and vegetables. Teucrium polium L. var. gnaphalodes (Lamiaceae), a wild-growing flowering plant that has many applications in food preparations and...

Baicalein (1), 6-hydroxyapigenin (6), 6-hydroxygalangin (13) and 6-hydroxy-kaempferol (14), which are naturally occurring flavonoids from a set of 14 hydroxy-flavones tested, exhibited high inhibitory effects on tyrosinase with respect to L-DOPA, while each of the 5,6,7-trihydroxyflavones 1, 6, 13 or 14 acted as a cofactor to monophenolase. Moreover, 6-hydroxykaempferol (14) showed the highest ...
